The castle originally stood near the River Dodder and was strategically placed in an area known as the Marches, which saw frequent conflicts involving local Gaelic clans. The castle was held by notable figures such as John Burnell and John Bathe, the latter serving as Attorney General and Chancellor of the Exchequer in Ireland during the late 16th century.

In the early 17th century, the castle was rebuilt by Pierse Archbold into a fortified baronial house with defensive turrets, reflecting architectural styles seen in the Scottish borderlands. Over time, ownership shifted among prominent families including the Loftus and Conolly families. By the 18th century, the castle was leased to the Quaker Ledwich family.

The surrounding area has since transformed with housing developments, but the historic significance of Knocklyon Castle remains tied to its role in defending the Pale and its association with influential Irish legal and political figures of its era.
